Binding Conditions / Buffer
Alternating Na+ and Li+ in the selection rounds, and both buffers contained 10 mMMg2+. In the end, a Mg2+-dependent aptamer was isolated to bind to UA.
The Na+ buffer contained 150 mM NaCl, while the Li+buffer contained 150 mM LiCl. Both buffers contained 20 mM Tris ,pH 8.3, 10 mM MgCl2and 10 mM KCl.

Refolding Protocol
If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target).
