Binding Conditions / Buffer
Using T4 PNK (Stratagene) for 45 min at 37C in PNK buffer (Stratagene).
In the presence of different concentrations of aptamers (0–1500 nm), human a-thrombin (2.5 NIH units per mL; CellSystems) and UFH (0.01 UmL^-1; Liquemin, Roche) were incubated in assay buffer (1x PBS, pH 7.4,
3 mm MgCl2, 1 mg mL^-1 BSA) for 5 min at room temperature in a total volume of 100 uL.
Binding Temp: 37°C
Refolding Protocol
If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target). Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding.
