Binding Conditions / Buffer

Binding buffer - (20 mM HEPES, pH 7.5, 150 mM NaCl, 2 mM MgCl2) High-affinity aptamers may fail to enrich if their hybridization to the capture strand is insufficiently stable under the selection condition. The authors suggest that a high salt condition could help and that they used 500 mM NaCl and 10 mM MgCl2 helped in raising the Tm enough that it prevented detachment from the capture strand on the beads.
