Binding Conditions / Buffer

Standard Reaction Conditions: 50 mM Tris-HCl, pH 7.5, 20 mM MgCl2 2X Gel Loading Buffer (equal volume used to quench reaction): 87 mM Tris base, 89 mM boric acid, 20% sucrose, 0.05% bromophenol blue, 0.05% xylene cyanol, 0.1% sodium dodecyl sulfate (SDS), 7.3 M urea, 1 mM EDTA. Additional 40 mM EDTA added.

Refolding Protocol

If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target).
