Binding Conditions / Buffer

For the first round of TECS-SELEX, the folded RNA was transferred to two 100-mm-dishes with 70% confluent TbRIII cells. After 10 min incubation at 37 °C, the RNA mixture was removed, and the cell surface was washed twice with 2 ml of buffer A, followed by the elution of the bound RNAs with 2 ml of EDTA-containing buffer (PBS-based cell dissociation buffer, Gibco/Invitrogen) at 37 °C for 5 min.

Refolding Protocol

If the oligo is a known aptamer sequence: For binding studies, perform a refolding protocol to ensure proper function (i.e. binding to antigen or target). Refer to the aptamer reference source for the appropriate refolding parameters and binding conditions. Note: it is unknown whether aptamer functions properly without refolding.
